Easy to prove by directly measuring the route with an altimeter.
The error can be anywhere from 50% to 200% depending on the route.
I don't think any club uses DeLorme for cumulative altitude gain.
At least for the centuries I've been on, the advertised gain is
pretty close to my altimeter's reading (much less than DeLorme).

I have seen lots of event rides that use DeLorme data. I rail against
it for two reasons; it scares off those afraid of climbing, and it
makes those who are willing to try a bit overconfident at times.
